I recommend to check your S2 MK3 preferences first - this is how they should look like:

If you are saying that nothing happens here when turning any of the two jog wheels:
then please make sure the S2 is connected, then open your Preferences -> Controller Manager, expand the Device menu and post a screenshot here. THX!
