There's a lot more that goes into it. First, I'm assuming the SSD being used for C: drive is internal. Are the other 3 drives internal or external USB plugins? Generally speaking you want to use internal drives if you have them because there's a speed advantage. USB external drives connected to USB C ports (given they're reasonably new and faster SSD) should be fine although not as fast as the internals typically.
Another consideration if you're using external SSDs is to make sure they're formatted NTSF as most of them come formatted exFAT to allow them to be used on Apple computers. But you pay a penalty in terms of stability and compatibility with many plugins if they're formatted exFAT on a PC.

All applications with their respective VST3 plugin versions should be installed to default folder on system drive… Then expansions and Kontakt libraries should go to Content on secondary drive…

Because i have update problems in Native Access
If these are update problems with expansions only then that is a known issue : Known Issue: Download Failed for updates on Windows (Expansions, Massive X Factory Library and more)
clear all the broken downloads.
reboot computer, open Native Access and go to tab Library
In tab Library locate something with tag Update in lower left
Use three dots in lower right to get choices and then select to uninstall product
wait for N.A. to update
then when choice install is present then re-install the product
Repeat above procedure for all products that needs updating OR wait for N.I. to fix the problem.
Please use latest version of Native Access for your OS .
Yes , you can change both the Download directory and the Content Directory in the N.A. File Management settings. However then please leave Applications path at the default.

Just to be crystal clear, the location where you install your individual addon products is determined in Native Access as shown above in @PoorFellow 's post. If you're asking about whether you can move the Native Access and Komplete Kontrol programs to a different drive location than your system drive, that's a much more complex situation. If you're initially installing them you might be okay to install them on a different internal drive connected directly to your motherboard. But there are a LOT of considerations here and you'd be MUCH better off describing your situation and the specifics about the computer your working with if you want to be sure about making the best choices.
Thanks to every one who answered my question. I have 4 SSD modules. One is C: with windows 11 on it. Also Native Instruments. The SSD is 4 TB but slowly because of all the applications it will be full. So i thought i want another SSD named I: for Native instruments. But if i understand the answer correctly it is not the good way to do it. It must be C: for the application.

Yes , applications and their plugin versions (standalone + VST3 , AU AAX) ought to be installed to default location on the system drive.
Then put all Content (Kontakt libraries and expansions , sample libraries) on a fast secondary SSD drive and also set Downloads to other than system for saving space.
All 4 are internal.I have a SSD special for applications. So if i have only install the Komplte Kontrol application on C: and the instruments on I: the problem is solved.
