Off the top of my head I would guess you have a microphone plugged into your Scarlet and you're hearing feedback. Or GR6 is set to use your Macbook speakers/microphone which would cause the same issue.

Mute the audio and check GR6 to make sure it's set to the correct audio interface and that the it's using the proper input.

It’s on the correct interface. Any other ideas? I’m extremely novice. I am a trumpet player who dables with guitar and am not good with tech. Thank tou!

Not really without knowing more about your setup. A screenshot of the audio preferences in guitar rig would be a good place to start. What's connected to your scarlet interface, and how is your scarlet set up (ie screenshot of the whatever the scarlet control software/mixer is)?

That sounds like feedback to me aswell, lower whatever speakers you're using to a minimum or move the mic away from them to test.

It's definitely not a feedback issue. I don't have a Mic set up. Even when I unplug the scarlet it happens. It's just a sound on my laptop itself. I changed preferences in Audio on my Mac for output to go through internal speakers to Scarlet. Either way I get this. It's a pause, then this screeching starts. Then pause again and keeps randomly doing this. If you can look at this attachment I sent, it happens when I hit either the right or left (or both) toggles on the "IN" section at the top of the screenshot. If I keep them muted I don't have the sound, but of course I need them on to play through GR. I said earlier it's GR6. Actually it's 5. I've used it for several years. Suddenly this started happening and I haven't changed anything that I'm aware of.

You say it's not a feedback issue but everything else you're saying points to feedback.

You say you don't have a mic set up, but you say the sound is on the laptop. Laptops have mics.

If you mute the inputs the sound stops. Again, points to feedback.

Turn off the laptop mic. Turn off the laptop speakers.

I don't know where the audio setup settings are in GR 5, but this is the screen I was looking for

In the screen shot you posted you can see that GR 5 is definitely getting input from something, so you need to check the audio device and the inputs and set them correctly.

Is it possible the built in sound card is note real clean and the high gain of some guitar rigs are picking it up. Open guitar cables will raise hell too. lol Really just mentioning external sound cards, they are generally isolated from computer noises being outside the machine. Most internal sound cards are pretty bad.
